The DTI FabSpin Blood DNA Kit II is designed to purify genomic DNA from whole blood. This kit employs a proprietary lysis buffer in combination with Spin Column membrane to efficiently purify genomic DNA from the biological sample. The protocol provides a simple method to achieve the rapid isolation of highly purified genomic DNA and the entire procedure can be accomplished within 30 mins. Genomic DNA obtained is suitable or a variety of applications, such as PCR, Southern blotting, RAPD, AFLP, RFLP and other molecular experiments.

Product Description: The DTI FabSpin Blood DNA Kit II is designed for rapid isolation of high purity DNA from whole blood of higher mammals, including humans with non-nucleated Red blood cells (RBCs). It can also be used for genomic DNA extraction from birds and fish containing nucleated RBCs.

High purity genomic DNA: Obtained with an A260/A280 ratio between 1.8-2.0 and concentration of 3-5 µg from healthy individuals.

Suitable for various applications: PCR, Southern blotting, RAPD, AFLP, RFLP, and more molecular experiments.

Compatible with EDTA, ACD, or heparin anti-coagulant blood samples: DNA purification can be done from buffy coat of human blood sample.

Storage and Shipment Conditions:

  1. Buffer components: Store at room temperature (15 – 25°C).
  2. Proteinase K (Lyophilised): Store at -20°C after receipt.
  3. Shipment temperature: Room temperature (15 – 25°C).

Materials required but not provided:
  1. 100% ethanol
  2. RNAse A (10 mg / mL) (as required)
  3. Centrifuge
  4. Vortex
  5. Water bath (56°C)
  6. Micropipettes (2-200 µl, 10-1000 µl)
  7. Pipette tips (200 µl, 1000 µl)
  8. Personnel protection equipment including labcoat, gloves, and goggles
  9. Proteinase-K to be purchased separately for catalogue numbers DT9702.25/ DT9702.100/ DT9702.250
Preparation Before Experiment:
  1. Proteinase K preparation: Dissolve the appropriate amount of lyophilised Proteinase K in the provided reconstitution buffer.
  2. Adjust water bath to 56°C.
  3. Before using the kit: Add the specified amount of 100% ethanol to Buffer WB and mix well.
  4. Pre-heat Elution Buffer/nuclease-free water to 65°C for improved elution efficiency.
